Effect of the proinflammatory cytokine IL-1β on the CP expression of NKCC1, pNKCC1 and pSPAK. Primary cultures were treated with either 1 pg/mL, 1 ng/mL, or 100 ng/mL IL-1β for 24 (A) or 48 (B) hours. Cells were lysed and subjected to immunoblotting for NKCC1, pNKCC1, pSPAK and proteasome 20 S on the same membrane. Molecular masses (shown in kDa) are indicated on the left. The NKCC1, pNKCC1, pSPAK and proteasome 20 S specific bands were quantified densiometrically. Bargraphs in (C) and (D) show the ratio of NKCC1/proteasome 20 S (left), pNKCC1/proteasome 20 S (middle), and pSPAK/proteasome 20 S (right) as mean ± SEM.
