Beta-catenin signaling in CNS ECs. In the brain Wnt7a/b binds specifically to the receptor complex Fzd4/Lrp5/Gpr124/Reck (left) and in the retina norrin binds specifically to Fzd4/Lrp5/Tspan12. Upon ligand binding the canonical Wnt signaling promotes the accumulation of beta-catenin in the cytoplasm and its translocation to the nucleus where it binds to TCF/LEF to promote the transcription of target genes that include genes promoting the induction and maintenance of the barrier
