The open reading frame of claudin-12 is located in exon 5 and is targeted by a BAC-based targeting vector that contains an expression and neomycin-selection cassettes. In the targeted allele, a deletion of 760 bps is present, which removes the entire coding sequence. Arrows indicate LoxP sites surrounding the selection cassette that are targeted by Cre recombinase. b Relative expression of different regions of the claudin-12 gene in heart muscle samples isolated from WT and claudin-12lacZ/lacZ C57BL/6J mice was assessed by qRT-PCR.
