LRP1shedding in human brain endothelial cells (HBMEC) following ADAM10 inhibition. HBMEC monolayers were exposed to human Aβ42 (2 μM) alone, Aβ42 (2 μM) and the ADAM10 inhibitor GI254023X (1 μM) for 48 h at 37 °C. LRP shedding was assessed by examining LRP content in the extracellular media using an LRP1 ELISA. Values represent mean ± SEM (n = 3) and are expressed as ng of extracellular LRP1 per ml of media. *p < 0.05 as determined by one-way ANOVA followed by Tukey’s post hoc analysis
